    Nemhem-vastartalmú modellek előállítása és gyakorlati alkalmazásai = Nonheme Iron-containing Enzyme Models and Their Possible Applications

    A jelen kutatási pályázat olyan nemhem-vastartalmú metalloenzimek (Oldható metán monooxigenáz, 1-amino-ciklopropán-1-karbonsav oxidáz, flavonol 2,4-dioxigenáz, pirokatechin dioxigenáz, szuperoxid diszmutáz) szintetikus modellezésére irányult, amelyek dioxigént használnak fel. A szerkezeti modellek előállításához - az irodalomban fellelhető ligandumok (pl. Salen) mellett - új háromfogú izoindolin-típusú ligandumokat állítottunk elő, majd komplexképződési reakcióik (Fe, Mn, Cu, Co, Ni) tanulmányozása után bioutánzó sztöchiometrikus és katalitikus oxidációs reakciókat dolgoztunk ki. A reakciókinetikai mérések és a reaktív intermedierek azonosítása alapján javaslatot tettünk a reakciók mechanizmusára. | The aim of this project was to work out structural- and functional models for nonheme iron-containing metalloenzymes which utilize dioxygen (Soluble methane monooxygenase, 1-aminocyclopropane-1-carboxylic acid oxidase, catechol dioxygenase, superoxide dismutase etc.) . The research project comprised the synthesis and characterization of new tridentate isoindoline-type ligands and their iron, copper, manganese, cobalt and nickel complexes, studies of their oxidation reactions (selectivity, intermediates, mechanism etc.), and testing them as suitable catalysts in various biomimetic reactions

    An acidophilic bacterial-archaeal-fungal ecosystem linked to formation of ferruginous crusts and stalactites.

    The presence of specialized microbial associations between populations of chemoautotrophic bacteria and archaea with ascomycetous fungi was observed inside stalactite-shaped mineral formations in a highly acidic cave environment. Metagenomic, chemical and electron microscopy analyses were used to investigate the relevance of these microbial ecosystems in the formation of stalactites. Ferric hydroxide produced by acidophilic bacteria and archaea was shown to be deposited onto fungal hyphae, resulting in complex mineralized stalactite-shaped structures. Thus, both archaeal-bacterial and fungal members of the ecosystem were shown to play an active role in the formation of stalactites

    Antimicrobial Activity of NCR Plant Peptides Strongly Depends on the Test Assays

    The symbiosis specific NCR247 and NCR335 cationic plant peptides of Medicago truncatula have been shown to exert antimicrobial activity against a wide range of microbes. However, their antimicrobial efficiency is clearly limited by divalent cations. Here, the antibacterial and antifungal activities of NCR247 and NCR335 peptides were compared to those of the well-characterized peptide antibiotics polymyxin B and the aminoglycoside streptomycin on three model microbes, Escherichia coli, Bacillus subtilis and Saccharomyces cerevisiae as representatives of Gram-negative and Gram-positive bacteria as well as eukaryotic fungi. The aim of the study was to assess how the killing efficiency of these peptides depends on various, widely used antimicrobial susceptibility assays. Validated resazurin microdilution assay was used to determine minimal growth inhibitory concentrations in three general test media (MHB, MHBII and low-salt medium LSM). Bactericidal/fungicidal activities were determined by the commonly used drop plate assay. The natural plant peptides showed distinct characteristics, NCR247 had a generally high sensitivity for Ca2+ and Mg2+ in the medium, while NCR335 proved to be a robust and strong antimicrobial agent with comparable efficiency values to polymyxin B. Activity data were confirmed visually, both NCR247 and NCR335 treatments at minimal bactericidal concentrations induced complete disruption of the membranes and provoked cell lysis on all tested microorganisms as observed by scanning electron microscopy

    Background/Aims: Congenital obstructive nephropathy (CON) is the main cause of pediatric chronic kidney diseases leading to renal fibrosis. High morbidity and limited treatment opportunities of CON urge the better understanding of the underlying molecular mechanisms. Methods: To identify the differentially expressed genes, microarray analysis was performed on the kidney samples of neonatal rats underwent unilateral ureteral obstruction (UUO). Microarray results were then validated by real-time RT-PCR and bioinformatics analysis was carried out to identify the relevant genes, functional groups and pathways involved in the pathomechanism of CON. Renal expression of matrix metalloproteinase (MMP)-12 and interleukin (IL)-24 were evaluated by real-time RT-PCR, flow cytometry and immunohistochemical analysis. Effect of the main profibrotic factors on the expression of MMP-12 and IL-24 was investigated on HK-2 and HEK-293 cell lines. Finally, the effect of IL-24 treatment on the expression of pro-inflammatory cytokines and MMPs were tested in vitro. Results: Microarray analysis revealed 880 transcripts showing >2.0-fold change following UUO, enriched mainly in immune response related processes. The most up-regulated genes were MMPs and members of IL-20 cytokine subfamily, including MMP-3, MMP-7, MMP-12, IL-19 and IL-24. We found that while TGF-β treatment inhibits the expression of MMP-12 and IL-24, H2O2 or PDGF-B treatment induce the epithelial expression of MMP-12. We demonstrated that IL-24 treatment decreases the expression of IL-6 and MMP-3 in the renal epithelial cells. Conclusions: This study provides an extensive view of UUO induced changes in the gene expression profile of the developing kidney and describes novel molecules, which may play significant role in the pathomechanism of CON

    MORFOMETRIAI KUTATÁSOK AZ EMBERI ARC TÖRTÉNETI EMBERTANI, IGAZSÁGÜGYI, ÉS GYÓGYÁSZATI CÉLÚ 3D REKONSTRUKCIÓJÁNAK FEJLESZTÉSÉHEZ = MORPHOMETRIC RESEARCH FOR DEVELOPING THE 3D FACIAL RECONSTRUCTION METHOD FROM THE PURPOSE OF HISTORIC- AND FORENSIC ANTHROPOLOGY, AND MEDICINE

    Létrehoztuk a FACE-R 3D koponya és- arc kutatói adatbázist, amely 425 élő vizsgálati személy és 5 kadáver fej adatait tartalmazza. Mysql adatbázisban tároljuk az adatállományokat, valamint a feldolgozás során nyert plusz információkat. Az adatbázis nemzetközi viszonylatban is egyedülálló, mert az illesztett 3D koponya CT-és arc modellek létrehozásával kiküszöböltük a fekvő helyzetben készülő gravitációs elmozdulást a CT-felvételeken. Előzetes elemzést végeztünk 60 egyén adatain 3D virtuális antropológiai és geometrikus morfometriai módszerekkel. Szignifikáns alaki összefüggéseket találtunk a koponya és az arc között. 20 egyén koponyáján feltáró EFA (Elliptic Fourier Analysis) elemzést végeztünk. Statisztikai módszerrel hiányzó adatpótlást végeztünk Szt. László király állkapocs rekonstrukciójához. A fekvő és- ülő helyzetű arcmodellek közti eltérés statisztikai vizsgálatára 3D biometriai elemzést végeztünk. Az arc matematikai modellezéséhez szükséges módszertani fejlesztések során standardizáltuk a 3D modelleket és programot írtunk a 3D felületi változékonyság jellemzésére, a min-max görbületi görbesereg meghatározására. Kidolgoztuk a koponya és az arc 3D „karakterisztikus geometriai modelljének” koncepcióját. Kísérleteket végeztünk a 3D külső orr rekonstrukció matematikai modelljének létrehozása. Elasztikus regisztrációs eljárást alkalmaztunk az arc modellezéséhez szükséges kiindulási adatok (3D template-ek) létrehozására. | We created the „FACE-R” 3D skull- and face research database, that is contain 425 living individuals’ and 5 cadavers’ head data. We stored the data files and the additional information in Mysql database. Thanks to the aligned 3D skull- and face models, we were able to eliminate the gravitational shift on CT-scans — recorded in supine position —, the database is unique in international relations as well. In the framework of the pilot study the virtual anthropological data collection and geometric morphometric analysis was performed on 60 persons’ data. We uncovered significant shape correlations between the skulls- and the faces. We performed exploratory EFA (Elliptic Fourier Analysis) on 20 persons’ skulls. Missing data complementation was accomplished applying statistical method for virtual mandible reconstruction of St. Laszlo, the Hungarian king. We carried on biometric analysis to uncover differences between the supine- and upright positioned 3D face models. In the framework of methodological development necessary for mathematical modelling the face, we standardized the 3D models than programmed an application for determine the min-max. curvature on the 3D skull- and face model surfaces. We worked out the concept of the „3D Characteristic Geometric Model” of the skulls and faces. We made some attempts to create a mathematical model for the 3D external nose reconstruction. We produced the 3D input templates for modelling the face applying „elastic registration” method
